The Francis Crick Institute

The Francis Crick Institute

The Francis Crick Institute (the Crick) is a research institute dedicated to discovering the fundamental biology underlying human health, with the aim of improving the diagnosis, treatment and prevention of human disease.

Summary

We seek a talented and motivated postdoc in the area of intestinal stem cell biology to join Dr. Vivian Li’s laboratory.

Dr Li’s laboratory focuses on understanding the mechanisms of intestinal stem cell lineage commitment and tumourigenesis. The ultimate goal is to translate the basic stem cell research to the clinic to aid cancer treatment and regenerative medicine.

Details of research projects currently being undertaken can be seen at: https://www.crick.ac.uk/vivian-li/.

Research techniques used in the laboratory include but not limited to: CRISPR/Cas9 gene editing, cell and 3D organoid culture, confocal imaging, molecular biology, protein biochemistry, generation and analysis of transgenic mice, histopathology, ChIP-seq and RNA-seq, scientific writing and presentation.

Project Scope/ Description

The project aims to characterise the regulatory mechanism of intestinal stem cell (ISC) lineage commitment. Previous expression analysis of ISCs and early progenitors in the lab has generated a handful of interesting candidates that potentially regulate ISC lineage commitment. To further characterise the functional roles of these genes, we aim to employ the recent CRISPR/Cas9 zygote-targeting approach to generate knock-in reporter mice to perform lineage tracing experiments and conditional knockout mice to characterise the function in the intestine. The result will help better understanding of the regulation of ISC lineage commitment, which may shield light on tissue regeneration and tumourigenesis.
