In the first part of my talk I will cover principles of cytokine production that can be used for objective ELISPOT analysis. I will cover reference sample strategies for scientific and regulated work, as well as serum free cryopreservation and testing strategies for assay maximization and standardization.  Data on the diagnostic value of granzyme B, perforin and TRAIL ELISPOT assays for human CD8 cell monitoring will be presented.
